Her mother. That's the main cause anyway. Ren said she would abort Akito if she wasn't raised as a male- 1. Because she thought it would be a disgrace for the most important zodiac (God) to be female, 2. (and most importantly) she thought a female child would replace her in Akira's (her husbands, Akito's father's) heart.
Akira died when Akito was young, and Akira was the only loving parent she had. After his death, Ren mentally abused Akito by calling her a worthless toy that she wished she never had, and that she was useless to everyone. This continued throughout Akito's life. Akito also had to take role as the leader of the family at a young age- and with that stress, the grief, the insecurities, and the abuse she recieved, she began to crumble.
She developed a fear of abandonment. She believed that the Zodiacs had a bond, and had to obey and stay with her- but when Yuki left to live with Shigure, Ren mocked Akito by saying that the bond "doesn't exist" and that they would all abandon her in the long run. Akito was obsessed with proving her wrong, and became very over protective of the zodiacs. Akito and Ren made a bet- allow the zodiacs to befriend "outsiders" and see if they'll come back- which is why she allowed Tohru to stay. She believed that love didn't exist outside the zodiac bonds, and she was going crazy trying to keep everyone with her- even if it meant being violent and cruel.
As time progressed, her fears, her inner termoil, her loneliness, and her dominating nature began to eat her away. She was desperate to keep everyone with her, and she was hurting them and herself in the process. She just became bitter and lost. ..I could go on and on, but I don' t want to spoil too much of the manga, nor do I want to lose room.
Which is why, she's my favorite character in Fruits Basket.
