Im thinking Vanessa lost hers to Nate because I remember her saying that she had wanted to be with Dan since she left and she didnt want to have sex wit him(so im guessing that means she wasn't with anyone else in vermont), and then the only person she had been with since the whole Dan thing is Nate.

I think Serena losted her virginity with Nate. They doesn't say in show with who she had lost it, but in the books she had her first time with Nate.

Serena didn't lose it to Nate, she lost it pre series. It's been said that she has slept with a LOT of guys, and since right after she slept with Nate, and then went to boarding school where she changed, it must mean she lost her virginity pre series.
