1. She slept with Chuck at the season finale.
2. The pregnantcy test was in her room.
3. In season 4 bloopers Blake says to Chuck "You're about to be a father.Wait, what? new storyline"
So i think it's Blair.

Okay the main character isn't pregnant. Plus there's already been pictures of Leighton filming and she dosent look pregnant at all. She would have a little bump by now if she was.

I think it's S because that would be maybe the reason she left to LA.
That would be the reason that she had a new job, the reason she changed.
And I think it would make S more mature and it would be interessant to see a single mom.

I don't think it's Charlie's because it would be very strange, and B has enough problems, and even if it was hers, I don't think she would be prepared to be a mother.

I don't think Ivy/Charlie has slept with anyone recently, so I don't think its her. And the GG producers have said that Serena is going to be very motivated and career-oriented, and I don't that'll be possible with a baby. Also, in a video of Leighton filming on-set, she says "I need to find a bathroom!" and runs off when Louis' sister asks if she wants a snack. By the way she says it, it sounds like she's about to throw-up. Finally, I don't think that we're going to see a baby bump on ANY of the characters, since that would completely give away who's pregnant.

I noticed that too. I don't think it matters if Ivy had sex with anyone, but maybe she was already pregnant when she got there?
The reason that I don't think Blair pregnant is because the writers wouldn't know how to handle a baby story line especially with Blair and her wedding ahead. Also, Blair is 20 years old. Turning 21.
Serena can't be preggo because surprisingly, she didn't have sex with anyone after 4x16.

Something I noticed that was weird with this question:
Why is Lily an option, lol. The pregnancy test was at the Waldorf's and she was under house arrest.
The only way this would make sense would be if they made the test a false positive and Lily actually turned out pregnant.

If Blair really was pregnant she either got an abortion (but this is the WB, so who are we kidding) or lost it by now. Leighton looks as slim as ever in filming picks, and by now Blair would be over 5 months along.

Also Blair slept with Loise maybe a week before she fucked Chuck, so without a paternity test, it would be impossible to tell who would be the father.

I think it's Eleanor, B or S getting knocked up would ruin the show. Ivy hadnt had sex with anybody had she?

@LoveMeABasshole actually this has nothing to do with Dair, and anyway they could still happen even if B was pregnant with Chuck's baby, i'm just pointing out the irony of it only being Chair fans that seem to want Blair pregnant, then i pointed out that i think it's sort of twisted to WANT a baby to be born into and unhappy, broken up relationship while the supposed mother is in another relationship in the hope of it saving the couple and people think it's who they want it to be, it's called wishful thinking, I for one don't want Blair to be pregnant because 1. she's not together with the supposed father. 2. that's not a good condition for a baby to be born into and 3. it'd probably ruin B's life and unlike a few people not naming any names i prefer to see her happy than depressed with a screwed up life and i'm not saying you don't have legit reasons to believe she's pregnant, i'm just saying i think it's kind of screwed up that you seem to want her to be, also i think it's kind of hypicrical for you to be telling me to grow up when you seem to think a baby's going to make your favourite couple reunite, also it's cool, you've yelled at me like five times before anyway.

Blair's way to smart to put a used pregnancy test exposed right at the top of the bin for the whole world to see, so my guess is that either Charlie or someone unimportant like that is pregnant or that phsyco Charlie/Ivy is trying to make it look like someone's pregnant as part of another one of her phsyo, wannabe clever and elaborate schemes.
